Trichoderma atroviride - investigations on regulation of expression of biocontrol related genes and strain improvement

Abstract

Trichoderma atroviride is a filamentous soil fungus which is known as an effective biocontrol agent against a wide range of economically important aerial and soil borne plant pathogens. Its mycoparasitic activity is due to the combination of successful nutrient competition, production of cell wall degrading enzymes and antibiosis. The use of the several strains of the genus Trichoderma as an alternative to chemical fungicides has received remarkable attention in recent years. However, the full-scale application of Trichoderma for biological control of plant pathogens has remained the exception because of their lower efficacy in biocontrol when compared with the respective chemical agents. The molecular mechanism of regulation of the T. atroviride endochitinase encoding gene ech42 was examined using EMSAs and reporter strains. Using these techniques a new DNA binding factor could be identified. To find out more about the role of another chitinase encoding gene (nag1) a disruptant strain was tested for its enzymatic activity and biocontrol abilities in vitro and in vivo. These experiments brought out the importance of nag1 in induction of the chitinolytic system of T. atroviride. Furthermore a heterologous expression system of the Aspergillus niger glucose oxidase was used to improve the plant protecting abilities of T. atroviride.

Trichoderma atroviride ist ein filamentöser Pilz im Boden, der als Biokontrollorganismus gegen eine Vielzahl von ökonomisch bedeutenden Pflanzenschädlingen eingesetzt warden kann. Seine antagonistische Wirkung beruht auf Nährstoffkompetition, Ausscheidung von Antibiotika und zellwandlytischen Enzymen. Die Chitinasen sind die bedeutendste Gruppe unter diesen zellwandabbauenden Enzymen. Zu näheren Untersuchungen der Regulation eines Endochitinase kodierenden Enzymes ech42 von T. atroviride wurden einerseits EMSAs herangezogen, andererseits wurden Glucose Oxidase Reporterstämme konstruiert. Mit Hilfe dieser Techniken konnte ein neuer DNA bindender Faktor in T. atroviride gefunden werden. Um mehr über die Bedeutung einer anderen Chitinase zu erfahren wurde ein nag1-negativer Stamm für enzymatische Messungenund Biokontrolluntersuchungen herangezogen. Die fehlende Expression dieser N-acetylglucosaminiodase wirkte sich auf das chitinolytische System von T. atroviride aus. In vitro wurde ein Rückgang der Chitinaseaktivitäten festgestellt, was auch zu einer deutlichen Reduktion der Biokontrollfähigkeit in situ führte. Ein Stamm, der die Aspergillus niger Glucose Oxidase unter Kontrolle des nag1 Promoters exprimiert, wurde auf eine mögliche Verbesserung der Biokontrolle untersucht. Dieser transgene T. atroviride Stamm zeigte deutliche Verbessuerungen im Pflanzenschutz bei hohen Pathogenkonzentrationen.
