Abstract

Protozoa are the cause of widespread morbidity and mortality in many countries of the world, including diseases such as malaria, sleeping sickness, Chagas disease, amoebosis and leishmaniosis. No prophylactic vaccines are available, many of the applied drugs cause serious adverse reactions and drug-resistance of parasites becomes a rigorous problem. Therefore, new anti-protozoal substances with an alternative mode of action are needed.

This project aimed to extract new natural compounds, focussing on bioactive substances occuring in tropical plant families, Asteraceae, Rutaceae, Meliaceae and Stemonaceae, respectively. The anti-plasmodial, anti-leishmanial and anti-trypanosomal in vitro activity of these plant-derived compounds was investigated in newly established assays aiming to find new drug candidates. Toxicity of the compounds was evaluated in human primary hepatocytes, the haemolytic activity was examined in freshly isolated erythrocytes.

Aglafoline showed 50% effective concentration (EC50) of 53.49 nM and rocaglamide showed an EC50 of 60.95 nM in fresh Plasmodium falciparum isolates. The anti-plasmodial activity was significantly below that of artemisinin with an EC50 of 15.63 nM, but it was moderately higher than that of quinine, which showed an EC50 of 180.75 nM. Aglafoline, rocaglamide, 5-hydroxynoracronycine, yukocitrine, and methyldambullin showed anti-leishmanial activity with EC50s of 6.99 µM, 5.76 µM, 4.42 µM, 0.88 µM and 1.10 µM, respectively compared to miltefosine with an EC50 of 1.27 µM. Methyldambullin, methylgerambullin and sakambullin showed promising, high anti-trypanosomal activities with EC50s of 1.7 µM, 1.23 µM and 5.18 µM, respectively compared to amphotericin B and benznidazole with EC50s of 0.71 µM and 30.89 µM, respectively.

The hepatotoxic activity of 5-hydroxynoracronycine, yukocitrine, methyldambullin methylgerambullin and sakambullin with EC50s of 201.87 µM, 147.06 µM, 48.43 µM, 41.77 µM and 37.09 µM, respectively was low compared to miltefosine, which showed an EC50 of 6.13 µM.

5-hydroxynoracronycine and yukocitrine were even less toxic than artesunate, which had an EC50 of 58.60 µM. The most toxic compounds were rocaglamide and aglafoline with EC50s of 4.94 µM and 5.07 µM, respectively. No haemolytic activity was obtained for these seven plant-derived compounds in concentrations between 150 µM and 1.56 µM, in contrast to miltefosine, where an EC50 of 119.03 µM was obtained.

In conclusion, seven compounds, namely the flavaglines aglafoline and rocaglamide, the acridones 5-hydroxynoracronycine and yukocitrine as well as the sulfur-containing amides methyldambullin, methylgerambullin and sakambullin showed significant anti-protozoal activity.

Zusammenfassung

Protozoen sind die Erreger für viele schwere Erkrankungen, die vor allem in tropischen Ländern zu Elend und Tod führen, wie z.B.

Malaria, Schlafkrankheit, Chagas-Krankheit oder die Leishmaniosen. Gegen diese Erkrankungen gibt es derzeit noch keinen Impfschutz, und viele der im Einsatz befindlichen Medikamente sind veraltet und weisen gefährliche Nebenwirkungen auf. Berücksichtigt man zusätzlich die weit verbreiteten Resistenzbildungen, so besteht ein großer Bedarf an neuen anti-protozoischen Substanzen mit alternativen Wirkmechanismen.

Die ungeheure Strukturvielfalt bioaktiver Naturstoffe vor allem in tropischen höheren Pflanzen, welche diese unter anderem als chemisches Verteidigungssystem gegen Mikroorganismen nutzen, bietet hier eine vielversprechende Quelle neuer Verbindungen. Das Ziel der vorliegenden Studie war, neue Naturstoffe aus verschiedenen tropischen Pflanzen, nämlich Vertreter der Asteraceae, Rutaceae, Meliaceae und Stemonaceae zu extrahieren und aufzureinigen. Weiters sollte das detaillierte Wirkprofil der selektierten Naturstoffe in breit angelegten in vitro Testreihen gegen Plasmodium falciparum, Leishmania infantum und Trypanosoma cruzi erfasst werden. Die Selektivität der Substanzen wurde in Toxizitätstests mit Normalzellen, nämlich Hepatozyten und Erythrozyten, ermittelt.

Sieben Substanzen, die Flavagline Aglafolin und Rocaglamid, die Acridone 5-Hydroxynoracronycin und Yukocitrin und die Schwefelamide Methyldambullin, Methylgerambullin und Sakambullin zeigten Wirkung gegen Protozoen. In mit Aglafolin und Rocaglamid behandelten Plasmodium falciparum-Isolaten von betroffenen Patienten, konnte eine 50% effektive Konzentration (EC50) von 53,49 nM bzw.60,95 nM erzielt werden. Die Substanz war ineffizienter verglichen mit Artemisinin, mit dem eine EC50 von 15,63 nM erreicht werden konnte, aber die Aktivität von Chinin mit einer EC50 von 180,75 nM konnte übertroffen werden. Aglafolin, Rocaglamid, 5-Hydroxynoracronycin, Yukocitrin und Methyldambullin zeigten Aktivität gegen Leishmania infantum Laborkulturen mit EC50 von 6,99 µM, 5,76 µM, 4,42 µM, 0,88 µM und 1,10 µM, im Vergleich zu Miltefosin, dem Standardtherapeutikum von Leishmaniosen, mit dem eine EC50 von 1,27 µM erzielt wurde. Methyldambullin, Methylgerambullin und Sakambullin zeigten vielversprechende Wirkung gegen Trypanosoma cruzi Kulturen mit EC50 von 1,7 µM, 1,23 µM und 5,18 µM, verglichen mit den Standardpräparaten, Amphotericin B und Benznidazol mit EC50 von 0,71 µM bzw. 30,89 µM.

Die Toxizität von 5-Hydroxynoracronycin, Yukocitrin, Methyldambullin, Methylgerambullin und Sakambullin gegenüber Hepatozyten mit EC50 von 201,87 µM, 147,06 µM, 48,43 µM, 41,77 µM und 37,09 µM war vergleichsweise gering zu Miltefosin mit einer EC50 von 6,13 µM.

5-Hydroxynoracronycin und Yukocitrin waren sogar weniger toxisch als Artesunat, mit welchem eine EC50 von 58,60 µM erzielt wurde. Es stellte sich heraus, dass Rocaglamid und Aglafolin mit EC50 über 4,94 µM und 5,07 µM relativ toxisch waren. Hervorzuheben ist dass in einem Konzentrationsbereich von 150 µM bis 1.56 µM mit keiner der sieben erfolgreich getesteten Substanzen eine hämolytische Aktivität beobachtet werden konnte. Hingegen wurde mit Miltefosin eine EC50 von 119,03 µM erreicht.
