Nach einer allgemeinen Einführung in die Thematik und einem kurzem Abriss aus derneueren Literatur, wird die Präparation von Cross-Linked Enzyme Aggregates (CLEAs) mitAlbumin als Additiv erfolgreich an den Lipasen Thermomyces lanuginosus (TL) und Lipase Baus Candida antarctica (CalB) demonstriert. Untersuchungen bezüglich des pH-Wertes vonGlutardialdehyd, der Albuminbeimengung und des Temperaturoptimums (TL: 45- 50 °C, CalB:75-80 °C) werden angestellt. Verglichen zur nicht quervernetzten Lipase liefert CalB diebesten Resultate bei einem Massenverhältnis von 1:16 Lipase/Albumin, welches zu einer24- fach gesteigerten Aktivität führt; TL bei einem Massenverhältnis von 1:12, welches zueiner 8-fach gesteigerten Aktivität führt. CLEAs aus Lipase TL erreichen bei der Umesterungvon Rapsöl mit 2,5 m% Katalysator 61 m% Ethylester innerhalb von 1,5 h Reaktionszeit. Beieinem Einsatz von 10 m% Katalysator wird ein Umsatz von 94 m% Ethylester erzielt. CLEAsaus CalB zeigen mit Novozym 435 vergleichbare Umsatzraten bei Glycerolyse- undEthanolysereaktionen. Innerhalb von 6 Rezyklierzyklen zeigen sich CLEAs aus TL undCLEAs aus CalB stabil (< 2 m% Produktabnahme pro eingesetzter Stunde). Für CLEAs ausTL und CalB wurden keine publizierten Daten für Umesterungsreaktionen gefunden. |

After a general introduction to the topic and a brief outline of the recent literature, thepreparation of Cross-Linked Enzyme Aggregates (CLEAs) with albumin as an additive isdemonstrated successfully with lipase Thermomyces lanuginosus (TL) and lipase B fromCandida antarctica (CalB). Investigations regarding the pH of glutardialdehyde, the albuminadmixture and the temperature optimum (TL: 45-50 °C, CalB: 75-80 °C) are employed.Compared to non-cross-linked lipase, CalB delivers the best results at a mass ratio of 1:16lipase/albumin, which leads to a 24-fold increased activity; TL at a mass ratio of 1:12, whichleads to a 8-fold increased activity. A transesterification yield of 61 wt% ethyl ester wasreached with 2.5 wt% CLEAs of lipase TL applied on rapeseed oil within 1,5 h reaction time.With 10 wt% of catalyst 94 wt% ethylester is achieved in the same time. CLEAs of CalB withNovozym 435 show comparable rates in ethanolysis and glycerolysis reaction. Within 6 cyclesof recycling CLEAs of TL and CLEAs of CalB remain stable (< 2 wt% decrease of product perinserted hour). For CLEAs from TL and CalB no published data were found fortransesterification reactions. |
