Gelée royale, der königliche Futtersaft der Bienen dient im Bienenstock der Fütterung von Larven und der Königin. Aufgrund seiner zahlreichen pharmakologischen Wirkun-gen wird Gelée royale in der Kosmetik und auch als Nahrungsergänzungsmittel einge-setzt. Bislang wurden Nachweismethoden für Pestizide in Pollen, Bienenbrot und ande-ren Bienenprodukten entwickelt, jedoch existiert keine publizierte Methode für den Nachweis in Gelée royale.In der vorliegenden Arbeit wurde eine Methode ausgearbeitet, um die Pestizide Dime-thoat (Organophosphat), Alpha-Cypermethrin (Pyrethroid), Clothianidin, Imidacloprid und Thiamethoxam (Neonikotinoide) in der komplexen Matrix des Gelée royale nach-zuweisen. Auf Grundlage von Literatur-basierenden Extraktionsverfahren wurden Me-thoden optimiert und mit Pestizid versetzte Proben mittels Hochdruckflüssigchromato-graphie und Liquid-Chromatographie-Massenspektroskopie analysiert.Als weiterer Schwerpunkt wurde in dieser Arbeit die Nachverfolgung von Clothianidin in der Honigbiene behandelt. Dazu wurde nach Fütterungsversuchen das Kopfgewebe, insbesondere die Hypopharynxdrüsen, sowie Honigmagen und Darm auf Clothianidin untersucht.Das Ergebnis der Analyse von Gelée royale ergab, dass die Neonikotinoide bei den verwendeten Extraktionsprotokollen allgemein gute Wiederfindungsraten ergaben und in Konzentrationen von 2 ng/mg Gelée royale in der HPLC detektiert werden konnten, wohingegen Alpha-Cypermethrin und Dimethoat deutlich schlechter nachweisbar wa-ren.Die Nachverfolgung von Clothianidin in Kopfgewebe, Honigmagen und Darm 6 Stun-den nach Fütterung von Clothianidin-hältiger Zuckerlösung an Ammenbienen ergab, dass Clothianidin mit den hier vorgestellten Methoden in diesen Geweben nicht nach-weisbar war. |

Royal jelly is produced by honeybees to feed larvae and the queen. It also is used in cosmetics and as food supplement due to its numerous pharmacological effects. Detec-tion methods for pesticides in pollen, bee bread and other bee products have been de-veloped, but so far, no method has been published for detection of pesticides in royal jelly.In this study a method has been developed to determine pesticides in complex matrices such as royal jelly. These pesticides were dimethoate (organophosphate), alpha-cypermethrin (pyrethroide), clothianidin, imidacloprid and thiamethoxam (neonico-tinoids). Based on established methods from literature extraction protocols have been optimized and samples of pesticide-spiked royal jelly have been analyzed by high pres-sure liquid chromatography and liquid chromatography-mass spectrometry.A further focus of this work was the tracking of clothianidin in honeybees. To address this topic, the head tissue, in particular the hypopharyngeal glands, honey stomach and intestine were examined for clothianidin after various feeding experiments.The result of the analysis of royal jelly showed that the neonicotinoids are also detecta-ble in concentrations of 2 ng/mg royal jelly. Alpha-cypermethrin and dimethoate are only detectable at higher concentrations.The result of the analysis of royal jelly showed that with the sample preparation method presented here, neonicotinoids show high recovery rates, whereas alpha-cypermethrin and dimethoate pose major problems in detection.Tracking of clothianidin in head tissue, honey stomac and midgut of clotianidin-fed nurse bees after 6 hours of intake revealed that clothianidin was not detectable in these tissues with the methods presented herein. |
